Yes, the Ohtani Rule is supported.
You can start a DH rule game as both the starting pitcher and the DH.
If you select the starting pitcher in [Batting Lineup Settings] and then select the batting order for the DH (the position with the [DH] icon), you can choose between [Starter + DH] and [Remove from DH].
